L'antiga ciutat de Tenos es trobava al sud-oest de l'illa al lloc on avui hi ha Aios Nikólaos, la capital actual. A la seva rodalia es trobava un temple dedicat a Posidó, situat en una cova, en el qual se celebraven festivals a què assistien gairebé tots els habitants de l'illa. Posidó apareix a les monedes de Tenos. Una altra ciutat de l'illa fou Eríston, a l'interior, on ara hi ha el poblet de Komi. Una curiositat de Tenos era una font l'aigua de la qual no es podia barrejar amb vi, que fou i es el producte principal de l'illa, on en destaca la malvasia, originalment sorgida a Monemvasia, al Peloponès.
Quant Xerxes va envair Grècia els habitants van haver d'acceptar el domini persa i servir al seu exèrcit. Un trirem de l'illa va desertar a la batalla de Salamina (480 aC) i, així, el nom de Tenos fou inscrit a Delfos entre els dels estats grecs que havien derrotat els perses. També van tenir el nom inscrit a l'estàtua de Zeus a Olímpia amb tots els estats que havien combatut a Platees. Posteriorment foren aliats d'Atenes, ciutat a la qual pagaven un tribut de 3.600 dracma|dracmes.
Alexandre de Feres va posseir l'illa per un temps, i després Marc Antoni la va cedir als Rodes|rodis.
Després de la conquesta de Constantinoble pels llatins, Tenos va passar a Venècia fins que la van cedir a l'Imperi Otomà arran de la pau de Passarowitz el 1718.
